Hematoxilina de Mayer la especie química activa es el complejo formado por hemateína (hematoxilina oxidada por yodato potásico) con sulfato de aluminio y potasio.
Este complejo tiene una carga positiva y por lo tanto es capaz de unirse a sitios aniónicos presentes en las proteínas histonas de la cromatina.
